Demanding excellent HDL?
Purified KALEN Human HDL (d=1.063 - 1.21 g/mL) is ultrafiltered through a membrane and packaged aseptically under nitrogen. The product is provided in a solution with 154 mM NaCl and 0.34 uM EDTA, at pH 7.2. The protein concentration is 10 mg/mL. The purity of the product is assessed by agarose gel electrophoresis.

Relative Electrophoretic Migration
To ensure that each lot of KALEN Human HDL has the same high quality, Kalen has implemented rigorous manufacturing protocols and stringent QC tests. The results depicted in the figure to the right demonstrate the consistency of KALEN Human HDL as determined by Relative Electrophoretic Mobility (REM). As shown, the variation in REM was minimal for the nine preparations tested. REM is calculated based on the migration of HDL compared with an LDL reference. Results are plotted as a percentage of the mean.
The HDL class of lipoproteins is one of the most dynamic and diverse. Two-dimensional gel electrophoresis can distinguish multiple subclasses. HDL particles contain Apo A-I, Apo A-II, Apo C, and Apo E; the exact protein composition depends on the nature of the HDL particle. The particles are 9-15 nm with a density of 1.063-1.210 g/mL. HDL is secreted from the liver in a nascent discoid form; they are poor in lipid and can be found in the pre-beta fraction following agarose gel electrophoresis. As the particles mature with the transfer of cholesterol mediated by lecithin-cholesterol acyl-transferase from tissues, they become spherical in shape. The mature form is found in the alpha migration fraction following agarose gel electrophoresis. HDL subclasses, HDL2a, HDL2b, and HDL3, are inter-converted by a variety of enzymes. Mature HDL particles return cholesterol esters to the liver by way of the SR-BI receptor.
